Buspirone is an antianxiety agent used to treat anxiety and manage generalized anxiety disorder. Noticeable improvements in mood usually appear after 2-4 weeks of daily use with a recommended trial period of 4 to 12 weeks. “The mechanism of action of buspirone is unknown. Buspirone differs from typical benzodiazepine anxiolytics in that it does not exert anticonvulsant or muscle relaxant effects. It also lacks the prominent sedative effect that is associated with more typical anxiolytics. In vitro preclinical studies have shown that buspirone has a high affinity for serotonin (5-HT1A) receptors. Buspirone has no significant affinity for benzodiazepine receptors and does not affect GABA binding in vitro or in vivo when tested in preclinical models. Buspirone has moderate affinity for brain D2-dopamine receptors. I agree that I:

am age 18 years old or older
have not had a hypersensitivity reaction to buspirone or any component of the formulation
have a diagnosis of generalized anxiety disorder and have already been taking buspirone as prescribed by an authorized healthcare provider
do not take a monoamine oxidase inhibitor (MAOI) such as isocarboxazid, phenelzine, tranylcypromine, or selegiline currently or within the past 14 day
will not initiate therapy with a MAOI until 14 days after discontinuation of buspirone
will avoid drinking large quantities of grapefruit juice while taking buspirone as this increases buspirone concentrations
am not receiving linezolid, intravenous methylene blue, or pimozide nor intend to while on buspirone
do not have a history of liver (hepatic) or kidney (renal) impairment
do not have a history of seizure disorder
am not pregnant or breastfeeding
Understand increased risk of serotonin syndrome when taken with another or multiple serotonergic drugs including selective serotonin reuptake inhibitors (SSRIs) (e.g. citalopram, escitalopram, fluoxetine, paroxetine, sertraline, fluvoxamine), serotonin-norepinephrine reuptake inhibitors (SNRIs) (e.g. venlafaxine, desvenlafaxine, duloxetine), or St. John’s Wort
Understand buspirone does not exhibit cross-tolerance with benzodiazepines or other sedative/hypnotic agents therefore, it will not block the withdrawal syndrome often seen with cessation of therapy with these drugs
have already been taking the initial doses (buspirone 5 mg and buspirone 7.5 mg) or have been previously prescribed buspirone 10 mg or buspirone 15 mg if selecting buspirone 10 mg or buspirone 15 mg.
Do not nor intend to take the following drugs concurrently with buspirone due to severe or major drug-drug interactions: linezolid, methylene blue, MAOIs, erythromycin, fuisdic acid, nefazodone, apalutamide, carbamazepine, encorafenib, enzalutamide, fosphenytoin, lumacaftor, ivacaftor, mitotane, phenobarbital, phenytoin, primidone, rifampin, adagrasib, atazanavir, ceritinib, clarithromycin, cobicistat, darunavir, idelalisib, indinavir, itraconazole, ketoconazole (systemic), levoketoconazole, lonafarnib, lopinavir, mifepristone, nelfinavir, nirmatrelvir, ombitasvir, paritaprevir, ritonavir, posaconazole, saquinavir, tucatinib, voriconazole
